IMMUNOADSORBENTS FOR THE TREATMENT OF EXPERIMENTAL
ALLERGIC ENCEPHALOMYELITIS IN GUINEA PIGS

The modern presumption of multiple sclerosis as a demyelinating disease of autoimmune nature contributes to the development of biotechnological approaches for removal of autoantibodies, immune complexes and specific clones of CD4 + T-lymphocytes from the patients systemic circulation for this disease treatment. Preclinical study of new efferent means conducted on laboratory animals with experimental allergic encephalomyelitis.

For the treatment of experimental allergic encephalomyelitis in guinea pigs, we used immunosorbents based on oxidized carbon granular matrix SCN and KAU with immobilized myelin basic protein MBP. Experimental allergic encephalomyelitis was induced by single subcutaneous injection of 100 ?g of MBP in complete Freund’s adjuvantt. Immunosorbents were prepared by chemical immobilization of MBP on SCM and KAU carbon granular matrices pre-oxidized with nitric acid in the reaction of covalent linking in the presence of water-soluble 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide. Synthesized immunosorbents were able to save up to 84% and 90% of the absorptive capacity for relatively small and medium molecular compounds of endogenous origin, respectively.

The development of experimental allergic encephalomyelitis of guinea pigs was accompanied by increasing in number of T-lymphocytes, high-level augmentation of anti-MBP antibodies and increased activity of acid phosphatase and trypsin-like proteases in blood serum. Immunosorption was carried out on the stage of experimental allergic encephalomyelitis manifestation or in latent period. It was found that synthesized immunosorbents were in therapeutic efficacy both being used in the latent period of disease and in the presence of its characteristic signs; they enabled to remove up to 37% anti-MBP antibodies from blood serum of guinea pigs with experimental allergic encephalomyelitis and to reduce activity of serum acid phosphatase by 54%.
