INDUCTION OF REGENERATION IN VITRO UNDER AGROBACTERIUM-MEDIATED TRANSFORMATION
OF SUNFLOWER INBRED LINE

The induction of regeneration in vitro of sunflower inbred lines (Helianthus annuus L.) under Agrobacterium-mediated transformation by strain LBA 4404 harboring antisense suppressor for proline dehydrogenase with gene neomycinphosphotransferase II were investigated. After cocultivation, significant decreasing of forthputting frequency (FF) under direct organogenesis out of a segment of seedling (half cotyledon, hypocotyl 1–2 mm size) 3–4-days after germinating was shown. It was found out that regeneration frequency depended on genotype as well as both selective, bactericide agents and ultrasound. On the whole, kanamycin (Kan, 100 mg/l) has shown negative effect, cefotaxime-positive effect (Cf, 500 mg/l) on FF while sonication (15 s, 44 kHz) increased significantly the realization of morphogenetic potential. It was proposed that procedure Kan- selection had to be done after lag-period of cultivation with bactericide agent.
