THE SPECIFIC INTERACTION PARAMETERS OF SERUM ANTIBODIES
WITH А RECOMBINANT ANALOGUE OF HERPES SIMPLEX VIRUS TYPE 2, GLYCOPROTEIN G

Specific interaction parameters such as affinity and avidity of serum IgG in the formation of immunocomplex with recombinant antigen of herpes simplex virus type 2 were determined.

It was found that all the herpes simplex virus type 2 positive sera contained high-affinity antibodies (the exponent of the affinity constant was in the range 107–108 M–1). The highest value of avidity index (81,15±11,15)% was observed in serum antibody number 2 while for the samples number 1 and number 3 these values were lower: (62,50±15,50)% and (65,0±15,10%) respectively, although there was not significant diffe rence between the amples (P > 0,05). The results of the avidity estimation for these three sera were in good agreement with theoretical values of affinity constants which were measured by the indirect ELISA according to the method of Freguet and others using the equations of Scatchard, Klotz and the equation of Stevens.

For one of the sera (№ 4) it was found the discrepancy between the affinity and avidity of antibody levels. Thus the affinity constant value calculated with the equation of Stevens was (2,08 ± 0,25)•108 M–1 and the avidity index (23,50 ± 6,50)% was significantly lower (P < 0,05) with respect to these three serums.

Obtained results are a contribution to the improvement of existing methods of laboratory diagnosis of herpes simplex virus and the detection of virus-specific antibodies in serum.
